HOW TRI-MIX WORKS

Tri-Mix is injected into the corpus cavernosum, where the three agents act on smooth muscle through separate mechanisms. Because they work directly on the tissue rather than amplifying an arousal signal, the response does not depend on the pathway oral medications require.
Alprostadil raises cAMP
Prostaglandin E1 relaxes smooth muscle through the cAMP pathway, independent of nitric oxide signaling.
Papaverine inhibits phosphodiesterases
Non-selective phosphodiesterase inhibition preserves both cGMP and cAMP, adding to the relaxation effect.
Phentolamine removes constriction
Blocking alpha-adrenergic receptors lifts the signal that keeps vascular smooth muscle contracted.
Dose is titrated in person
