Enclomiphene is the trans-isomer of clomiphene and acts as an estrogen receptor antagonist in the hypothalamus and pituitary. Because estrogen is part of how the body decides it has enough testosterone, blocking that signal prompts the pituitary to release more LH and FSH, which drives the testes to produce more testosterone directly.

Enclomiphene blocks estrogen receptors in the hypothalamic-pituitary axis, reducing the feedback that tells the body to slow testosterone production.
